Seamlessly Expanding a Randomized Phase I 1 Trial to Phase I 11

چکیده

A sequential Bayesian phase II/III design is proposed for comparative clinical trials. The design is based on both survival time and discrete early events that may be related to survival and assumes a parametric mixture model. Phase I1 involves a small number of centers. Patients are randomized between treatments throughout, and sequential decisions are based on predictive probabilities of concluding superiority of the experimental treatment. Whether to stop early, continue, or shift into phase I11 is assessed repeatedly in phase 11. Phase I11 begins when additional institutions are incorporated into the ongoing phase I1 trial. Simulation studies in the context of a nonsmall-cell lung cancer trial indicate that the proposed method maintains overall size and power while usually requiring substantially smaller sample size and shorter trial duration when compared with conventional group-sequential phase I11 designs.

Topical Channa striatus 5% cream for inflammatory skin conditions: A phase I randomized double-blind, controlled trial

Background: Channa striatus or better known as the haruan fish is an indigenous, snakehead, striped, air breathing freshwater fish of Malaysia and has long been consumed traditionally during post-operative and post-partum period to speed recovery. Most medical properties of the haruan are contributed to its high content of essential fatty and amino acids. This was a phase I clinical trial to ex...

A phase I/II clinical trial for adult recurrent glioma using 131i-tm-601, an iodinated peptide derived from scorpion venom

131I-TM-601 is a 36-amino acid peptide, called chlorotoxin (TM-601), derived from scorpion venom labeled with I-131. TM-601 binds a receptor on the surface of tumor cells, and not on normal cells. A single dose of 131I-TM-601 administered intracranially to human xenografted mouse models of glioma has been shown to extend survival up to 269% in multiple studies. 131I-TM-601 is in a multi-center ...

Commentary: Why we should report results from clinical trial pilot studies

Should clinical trial pilot studies be reported? Thebane, Ma, Chu, et al. argue in their discussion of the conduct of pilot studies, that they should be published [1]. Others, such as the editorialist for The Diabetes Educator, take a more cautious approach to publishing pilot studies [2]. I agree with those who advocate publication. I might also note that a September 27, 2012 search of “random...

Numerical Study of Single Phase/Two-Phase Models for Nanofluid Forced Convection and Pressure Drop in a Turbulence Pipe Flow

In this paper, the problem of turbulent forced convection flow of water- alumina nanofluid in a uniformly heated pipe has been thoroughly investigated. In numerical study, single and two-phase models have been used. In single-phase modeling of nanofluid, thermal and flow properties of nanofluid have been considered to be dependent on temperature and volume fraction. Effects of volume fraction a...
